- 1502: Lucas Vazquez de Ayllon heard the stories about the New World joined the expedition to the West Indies led by Nicolas de Ovando with Diego Velazquez. A fellow adventurer was Hernando Cortes
- Lucas Vazquez de Ayllon did well in the New World and was appointed a member of the Superior Council in San Domingo
- Due to his connection with Hernando Cortes it is quite possible that he accompanied him on his exploration of Mexico and the conquest of the Aztec Empire ( although this is not confirmed

- 1521: He sent an expedition to Florida under Francisco Gordillo in a quest to find the Northwest passage. The expedition landed near Cape Fear in North Carolina in June 1521
- 1524: Lucas Vazquez de Ayllon followed the Gordillo expedition and discovered Chesapeake Bay and the James River
- 1524: Lucas Vazquez de Ayllon reports to King Charles V of his his discoveries resulted in De Ayllon being granted the land he had discovered
- 1526: Lucas Vazquez de Ayllon leads a new expedition of 2 ships and 600 colonists and founded San Miguel de Guandape (near Jamestown)
- The colony of San Miguel de Guandape has a notorious place in history as the first colony to use negro slaves in the United States
- 1526: Lucas Vazquez de Ayllon died 18 October, 1526 of a fever
